LC-Raman spectrometer

The combination of high performance liquid chromatography (HPLC) with Raman spectroscopy achieves the on-line separation and accurate identification to the eluent.

We succeeded in combining the HPLC (PU-4180-LPG, JASCO) system equipped with a C18 reversed phase column (5C18-AR-300, Cosmosil) with the Raman spectrometer (Ventana 785, Ocean Optics) with the aid of the vertical flow method that improves the signal collection efficiency.

The vertical laminar flow of the HPLC eluate is formed through a pinhole (100-200 um) on the polyimide film (50 um thickness). 785 nm cw laser beam is coupled to an optical fiberand focused on the pinhole with an objective lens of a Raman probe. The beam excites the Raman scattering in the laminar flow. The total reflection at the sample-air interface confines the excitation beam and the Raman signal, thereby improving the signal collection efficiency.
